Abstract
Objective:To observe the synergistic effect of hyperthrnia and cisplatin on human lung adenocarcinoma cell line H1299.Methods:The cells treated 42 degree for 2 hours or not were detected cisplatin sensitivity with CCK-8 assay. After 42 degree for 2 hours, ciplatin 10μg/ml for 24 hours or the combination with both sides, the inhibition ratio was detected wtih CCK-8 assay and flow cytometry analyzed DNA contents in every phase of cell proliferation, and we observed their synergistic effect on cytotoxicity and cell cycel.Results:After hyperthermia, the IC_ 50 values to cisplatin decreased from 9.51μg/ml to 6.07μg/ml. CCK-8 assay showed hyperthermia and ciplatin had synergistic cell killing effect, and FCM also showed hyperthermia had significant effect on cell cycle with increase of G0/G1 phase population and reduction of S phase population.Conclusion:Hyperthermia can significantly increase the sensitivity of H1299 cells to cisplatin.
