Abstract
Objective To determine the apoptotic effect and its mechanism of sinoactinomycin on the human CNE-2 cells of nasopharyngeal carcinoma in vitro. Methods CNE-2 cells were treated with sinoactinomycin at different concentrations. The growth inhibition effect of sinoactinomycin was determined by MTT assay.Apoptosis was evaluated under inverted microscope.Bax mRNA expression was detected by reverse transcriptase-polymerase chain reaction(RT-PCR).Results Sinoactinomycin significantly inhibited the growth of the CNE-2 cells. The Bax mRNA level was significantly increased in sinoactinomycin treated cells compared to non-treated cells in a concentration dependant manner. Conclusion Sinoactinomycin inhibits the growth of CNE-2 cells due to apoptosis induced by the up-regulatoin of Bax.
