Abstract

Objective To observe the effect of human nerve growth factor(hNGF) on UDP-Galactose:Ceramide Galactosyltransferase mRNA (CGTmRNA) in sciatic nervers of experimental diabetic polyneuropathy rats.Methods Diabetic rat was induced by a single intraperitoneal injection of streptozotocin.After 4 weeks of diabetes,hNGF(90U/kg/d) was injected intraperitoneal for 4weeks.The level of CGTmRNA,NGFmRNA and protein expressions in sciatic nerve were measured by RT-PCR and immunohistochemistry techniques.Results After 8 weeks of diabetes,CGTmRNA expression was increased significantly, NGFmRNA and protein levels were decreased in sciatic nerves.After treatment withhNGF for 4 weeks,CGTmRNA expression was decreased,and NGFmRNA and protein levels wereincreased.Conclusion Administration of hNGF can reverse abnormal expression of CGTmRNA in sciatic nervers of experimental diabetic polyneuropathy rats.
