Abstract
Aim:To observe the effect of human nerve growth factor(hNGF) on CalpainⅡ expression in sciatic nerves of experimental diabetic polyneuropathy rats. Methods:Diabetic rat was induced by a single intraperitoneally injection of streptozocin. After 4 weeks of diabetes,hNGF 90 U/(kg·d) was injected intraperitoneally for 4 weeks. The level of calpainⅡmRNA and protein expressions in sciatic nerves were measured by in situ hybridization and immunohistochemistry techniques.Results:After 8 weeks of diabetes, both calpainⅡmRNA and calpainⅡprotein expressions were decreased significantly. Treatment with hNGF for 4 weeks, calpainⅡmRNA and protein expressions were increased.Conclusion:Administration of hNGF can partly reverse abnormal expression of calpainⅡ in sciatic nerves of experimental diabetic polyneuropathy rats.
