Effect of TPM on Fas expression in hippocampus in PTZ induced chronic kindling seizure rats

Abstract

Objective To explore the effect of TPM on Fas expression in hippocampus in pentylenetetrazole(PTZ)induced chronic kindling seizure rats and the protection on neuron of hippocampus.Methods 48 rats were divided into normal control group(group A)、PTZ kindling epilepsy group(group B)、TPM 40 mg/kg group(group C)and TPM 80 mg/kg group(group D).Use chronic model of epilepsy induced by PTZ intraperitoneal injection.Changes of behavior,EEG recording and hippocampal histology were observed,and Fas expression was determined with the immunohistochemistry method.Results Compared with group B,the seizure activity and its degree decreased significantly(P0.01).The expression of Fas and apoptosis significantly increased in group B and TPM could reduce the expression of Fas and apoptosis(P0.01).Conclusion In PTZ induced chronic kindling rats,TPM could down-regulate the Fas expression in hippocampus,thus decreasing neuron apoptosis.
