Abstract
Objective To investigate the regulation effect of interleukin-1β (IL-1β) on the expression of VEGF in A549 cells. Methods A549 cells were incubated with IL-1β in the presence or absence of Celecoxib. After incubation, cells were harvested to detect VEGF mRNA by reverse transcription-polymerase chain reaction (RT-PCR), and the supernatant was collected to determine the concentration of VEGF protein by enzyme-linked immunosorbent assay (ELISA). Results After treatment with IL-1β, the expression of VEGF mRNA and protein in A549 cells were significantly higher than that in the control group(P0.05); IL-1β up-regulated VEGF mRNA and protein expression in a dose-dependent and time-dependent manner in A549 cells, after incubation for 24 hours, VEGF expression was increased. After Celecoxib intervention, VEGF expression decreased as compared with that of IL-1β induced group, the difference was statistically significant(P0.05); with the concentration of Celecoxib stepping up, VEGF expression was decreased in dose-dependent manner and the difference was significant (P0.05).Conclusion IL-1β upregulates VEGF expression in A549 cells,the mechanism may be at transcriptional level; Celecoxib down-regulates IL-1β induces VEGF expression in A549 cells.
