Abstract
AFLP analytic method was tested in the forty-two cultivars of Cymbidium sinense cultivars,originated from Guangdong,Fujian,and Taiwan provinces of China.Genomic DNA was extracted from the green leaves of Cymbidium sinense cultivars by the method of CTAB 9 pairs reproducible primers were selected from 64 pairs of primers.A total of 1 384 bands were observed,of which 1 333 bands(96.3%) were polymorphic.Each pair of primers produced 154 bands following amplification.The AFLP markers indicated that all cultivars were of different cultivar.39 cultivars had their specific bands or deleted bands.The Dice coefficients amongst the cultivars ranged from 0.422 2 to 0.824 5,with an average value at 0.611 2.The polymorphic ratio of C.sinense 'Da Mo' with sport mutations is 76.6%.The Dice coefficients amongst the cultivars ranged from 0.616 4 to 0.824 5,with an average value at 0.725 6.According to the dendrogram using unweighted pair group method using arithmetic averages(UPGMA),the cultivars were grouped into three main clusters.Cultivars with the same origin could be clustered together.
