Abstract
Objective To explore the effects of advanced glycosylation end products on the mRNA expression of connective tissue growth factor(CTGF) in rat mesangial cells. Methods Rat mesangial cells were cultured in vitro under standard conditions. After stimulated by AGE-BSA and BSA, respectively, RNA was extracted and TGF-β 1 and CTGF mRNA were assessed by RT-PCR. Results TGF-β 1 and CTGF mRNA were significantly upregulated by AGE-BSA in time- and dose-dependent manner compared with control BSA. AGE-BSA induced TGF-β 1 mRNA expression earlier than CTGF mRNA. Conclusion AGEs can significantly induce TGF-β 1 and CTGF mRNA expressions in cultured mesangial cells in time- and dose-dependent manner.
