Abstract
Objective To observe the protective mechanism of bone marrow mesenchymal stem cells on endotoxin-induced acute lung injury.Methods 8 healthy 4-month-old SPF grade of C57BL/6J mice were used for bone marrow mesenchymal stem cells isolated and cultured;90 male SPF grade SD rats were randomly divided into: normal control group(A group);LPS group(B group),bone marrow mesenchymal stem cells in high-dose group(C group),bone marrow mesenchymal stem cells in the low dose group(D group),bone marrow mesenchymal stem cells in the control group(E group).6,24,72 hours after modeling,the animals were killed at each time point 6.Lung tissue were collected,measured lung wet weight/dry weight.Lung tissue superoxide dismutase(SOD) activity and malondialdehyde(MDA) and myeloperoxidase(MPO) content.Pathological changes of lung tissue.Results LPS 24-hour group one dead,each time point two bone marrow mesenchymal stem cell therapy group compared with model group,the lung tissue SOD activity was significantly higher,MDA and MPO were significantly lower(P0.05);pathology significantly improved and lung wet weight/dry weight reduced.There was no significant difference between the normal control group and bone marrow mesenchymal stem cells group on these indicators.Conclusion Bone marrow mesenchymal stem cells has a protective effect on endotoxin induced acute lung injury,its mechanism Involve in antioxidant capacity.There was no significant damage in different species of bone marrow mesenchymal stem cell transplantation.
