Effects of chinese herbs compound Qishenyiqi on expression of I、III type of collagens in the fibrotic liver induced by DMN in rats

Abstract

Objective:To investigate the effect of Chinese herbs compound Qishenyiqi on expressions of Ⅰ、Ⅲ type of collagens in the fibrotic liver.Methods:Wistar rats were randomly divided into normal control group, model group, Chinese herbs compound Qishenyiqi intervened group, model control group, and Qishenyiqi therapeutic group. The animal models of liver fibrosis were induced by intraperitoneally injecting with dimethyenitrosamine (DMN). Liver fibrosis stage were evaluated on the liver tissue sections which were performed with HE and Reticulation's stain. Semi-quantitive RT-PCR were perfromed to determined mRNA levels of collagen type Ⅰand Ⅲ in liver tissues. Immunohistochemical staining were performed to show the positive expression of collagen type Ⅰand Ⅲ in liver tissues. Results: Compared with model group, liver fibrosis stage in Qishenyiqi intervened group were obviously lowered, and compared with model control group, the liver fibrosis were significanlty relieved after Qishenyiqi's therayp. In intervened and therapeutic groups, the liver tissue's mRNA levels of collagen type Ⅰand Ⅲ were all obviously lower than in the model and model control groups respectively. In intervened and therapeutic groups, the positive expression in liver tissues of collagen tpye Ⅰand Ⅲ were all much weaker than in the model and model control groups respectively. Conclusion: Chinese herbs compound Qishenyiqi has the positively preventive and therapeutic effects on the experimental liver fibrosis induced by DMN in rats, inhibiting the expression of collagen tpye Ⅰand Ⅲ liver tissues amy be one of the anit-fibrosis mechanisms.
