Expression of survivin in prostate cancer cell lines

Abstract
Objective: To study the expression of survivin in prostate cancer cell lines PC-3,PC-3M,DU145,LNCaP and 22RV1.Methods: The expression of survivin were investigated by western blot, RT-PCR and immunohistochemistry methods. Results: The results of immunohistochemical staining showed that the rate of positive cells of survivin were 23% in PC-3,18% in DU145,15% in LNCaP,11% in 22RV1 and 12% in PC-3M. Immunoscores for PC-3,PC-3M,DU145,LNCaP and 22RV1 were 49.42+4.71(±s),28.45+6.82,34.27+6.08,34.09+5.22 and 31.66+7.13. The highest expression of survivin was observed in the PC-3 cells compared with PC-3M,DU145,LNCaP and 22RV1 (F=22.84, P0.01). Western blot analysis of prostate cancer cell lines showed that the expressions of survivin were higher in PC-3,PC-3M and DU145 cells, but lower in 22RV1 and PC-3M cells. The RT-PCR analysis showed that the mRNA of survivin were higher in PC-3 and LNCaP cells but lower in the other prostate cancer cells(P0.05).Conclusion: The different expression of survivin were observed in PC-3、PC-3M、DU145、LNCaP and 22RV1 cell lines.
