Abstract
Objective To investigate the protective effect and mechanism of Xingnaoyizhi capsule on learning and memory acquisition impairment in mice.Methods Sixty KM mice were randomly divide into 6 groups: normal control group,model group,Huperzine- A group,Xingnaoyizhi capsule of high dose group,middle dose group and low dose group( 179.2,89.6,44.8 mg ·kg- 1).Learning and memory acquisition impairment models were induced by scopolamine hydrobromide intraperitoneal injection and assessed with step- down test.The levels of acetylcholine esterase( Ach E),choline acetyl transferase( Ch AT) and glutathione peroxidase( GSH- Px) in brain tissues were assayed by spectrophotometry; The pathological changes of brain tissue were observed by hematoxylin and eosin( HE) staining.Results Compared with model group,high dose group prolonged the latency periods of memory and significantly reduced the error times within 5 mins in impairment groups( P 0.05),the level of Ach E in high dose group was decreased,while the levels of Ch AT and GSH- Px were higher than that in the model group( P 0.05).The content of Ch AT in middle dose group was significantly increased( P 0.05),the level of Ach E and GSH- Px were changed compared with model group,but without significance.Brain tissues stained with HE showed that xingnaoyizhi capsule could remarkably relieve cerebral injury.Conclusion Xingnaoyizhi capsule has protective effect on learning and memory acquisition impairment in mice,which may be related to protect the central cholinergic nerve system and antioxidant effects.
