Abstract
Objective To investigate the influence of Edaravone combined with Nimodipine on the expression of vascular endothelial growth factor(VEGF) in rats with cerebral ischemia.Methods Ninety rats were randomly divided into 5 groups: normal control group,cerebral ischemia group,Edaravone group,Nimodipine group and Edaravone+Nimodipine(EN) group,and each group was divided into the ischemia 12 h,24 h,48 h sub-groups.The cerebral ischemia model was established by middle cerebral artery occlusion(MCAO) intraluminal suture method.One h after the moldel made,the rats in each intervention group were intraperitoneal injected Edaravone 3 mg/kg or Nimodipine 2 mg/kg or Edaravone 3 mg/kg+Nimodipine 2 mg/kg,respectively;the normal control group and cerebral ischemia group were intraperitoneal injected normal saline.The rats was sacrificed at specific time points.The expressin of VEGF mRNA in brain tissue was detected with reverse transcription-polymerase chain reaction(RT-PCR).Results Compared with normal control group,the expression of VEGF mRNA in brain tissue in cerebral ischemia 24 h,48 h sub-groups of each group were significantly lower(all P0.01),but which in each intervention group were significantly higher than that in the cerebral ischemia group(all P0.01).While,which in EN group was significantly higher than Edaravone group and Nimodipine group respectively(all P0.01).Conclusion Edaravone and Nimodipine can up regulated the expression of VEGF mRNA in ischemia brain tissue,and combination of the two will be better.
