Expression of IFN-γ,IL-10mRNA in peripheral blood mononuclear cells from patients with Hepatocellular Carcinoma

Abstract

Objective:To analyze the expression levels of interferon gamma (IFN-γ) and interleukin 10 (IL-10) mRNA in peripheral blood mononuclear cells (PBMCs) in the pathogenesis of hepatocellular carcinoma (HCC).Methods:PBMCs were isolated from 20 patients with HCC and the total RNA was extracted.The expression levels of IFN-γ and IL-10 mRNA in PBMCs were amplified by using semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR).The density of electrophoresis band in PCR products was scanned by gel scanning system.The results were compared with that from normal healthy volunteers.Results:RT-PCR analysis showed that the expression levels of IL-10 mRNA were significantly increased in patients with HCC compared with that in healthy individuals (P0.01).Whereas IFN-γ mRNA levels were significantly decreased compared with that in healthy individuals (P0.01).Conclusion:Th1/Th2 is imbalance in PBMCs with HCC.
