Effects of Ginsenoside Rb_1 on Ventricular Remodeling in Rats with Acute Myocardial Infarction

Abstract

Objective To investigate effects of Ginsenoside Rb1 on left ventricular(LV) remodeling in rats with acute myocardial infraction(AMI).Methods Fifteen male Sprague Dawley rats surviving AMI,which was achieved by ligating coronary artery,were randomized to AMI control group (n=7) or Ginsenoside Rb1 treatment group(n=8).Sham-operated rats(n=8) were selected randomly.After surgery,treatment of Ginsenoside Rb1(4mg·kg~(-1)·d~(-1),for treatment group) or saline(for AMI control and sham-operated groups) was started by intraperitoneal injection.After 4 weeks of treatment,hemodynamic studies and morphological analysis were performed and the collagen Ⅰ of LV non-infarct zone were detected with immunohistochemical staining.Results Compared with Sham-operated rats,ratio of left ventricular weight to body weight(LVW/BW),left ventricular diameter(LVD),type Ⅰ collagen content and the LV end diastolic pressure(LVEDP) in AMI control group were significantly increased(P0.01),while the LV systolic pressure(LVSP),the maximum rising and dropping rates of LV pressure in AMI control group were significantly reduced(P0.01).In contract,compared with AMI control group,LVW/BW,LVD,type Ⅰ collagen content,LVEDP and size of MI in Ginsenoside Rb1 treatment group were significantly decreased(P0.01 or 0.05).While LVSP and the maximum rising and dropping rates of LV pressure in AMI were significantly increased(P0.05).There were no significant difference on the heart rate(HR) among the three groups.Conclusion Ginsenoside Rb1 can prevent and treat LV remodeling after AMI in rats efficiently.
