Abstract
Objective To study the effect of learning and memory and the protein expression of β-AP in the hippocampus in AD model rats,provide a theoretical basis for the clinical application of acupuncture treatment on AD. Methods 50 healthy male Wistar rats,after do behavioral test,30 selected rats were randomly divided into normal group,model group and EA group.Modeling method was used microinjection of each side of the lateral ventricle of 10% in streptozotocin 10 μL.To stimulate the Baihui,Dazhui,Zusanli acupoint in EA group.To detect learning and memory ability using the Morris water maze and detect beta-AP-positive cells expression in the hippocampus with immunohistochemical staining. Results The learning and memory ability in EA group was significantly increased.The escape latency and swim distance was significantly shorter(P0.01).It was significant increase in the percentage of the original platform swimming time in whole swimming time.The first time through the original platform location was significantly shorter(P0.01).beta-AP protein expression levels of Hippocampus in model group increased than normal group(P0.01).Compared with model group,beta-AP protein levels decreased after treatment in EA group(P0.01). Conclusion After EA treatment,learning and memory ability significantly improved in AD rat.Acupuncture can reduce beta-AP protein levels within the cells of the AD hippocampus of rats.Acupuncture has a therapeutic role in AD rats.
