Application of automatic hematology analyzer Sysmex XT-4000i in cells counting for ascite fluid

Abstract
Objective To evaluate the application of body fluid mode in automatic hematology analyzer Sysmex XT-4000i (RET channel) in ascite fluid cells counting. Methods One hundred fresh ascite fluid specimens were used for cell counting by automatic hematology analyzer Sysmex XT-4000i (automatic method) and improved Neubauer counting board (manual method) respectively. The results were compared by statistics. Results A good correlation was found between automatic method and manual method. There was no significant difference of the results in leukocyte[(1.403±5.527)×109/L vs. (0.858±2.740) ×109/L], mononuclear cells[(0.430±1.172) ×109/L vs. (0.343±0.832) ×109/L] and multinucleated cells[(0.969±5.011) ×109/L vs. (0.515±2.371) ×109/L] counting by automatic method and manual method (P 0.05). The coefficient variation(CV%) of the automatic method was higher than manual method.The carry over rates of total cells, erythrocytes, leukocytes, mononuclear cells were 0.037%, 0.000%, 0.161%, 0.070%, while the carry over rates of multinucleated cells was 5.633%. Conclusion Sysmex XT-4000i can be applied in cells counting for routinely clinical ascite fluid detection.
