Abstract

Objective:To observe the effect of estradiol combined with different doses of puerarin on osteoporosis of ovariectomized rats,provide a experimental basis for finding out the optimal dose of puerarin combined with estradiol to treat type Ⅰ osteoporosis.Methods:70 5-month-old healthy female rats were divided into 7 experimental groups,10 rats in each group,the models of ovariectomized rats were established in different groups except sham-operated group.The rats in puerarin-50 group were treated with subcutaneous injection of puerarin(50 mg/kg,once a day);the rats in estradiol-200 group were treated with subcutaneous injection of estradiol(200 μg/kg,twice a week);the rats in estradiol+puerarin 10 group were treated with subcutaneous injection of estradiol(100 μg/kg,twice a week) and puerarin(10 mg/kg,once a day);the rats in estradiol+puerarin 25 group were treated with subcutaneous injection of estradiol(100 μg/kg,twice a week) and puerarin(25 mg/kg,once a day);the rats in estradiol+puerarin 40 group were treated with subcutaneous injection of estradiol(100 μg/kg,twice a week) and puerarin(40 mg/kg,once a day).5 rats were selected from each group randomly at 10 and 20 weeks after treatment,then their left thighbone sections were obtained to observe bone tissue,and their right thighbone specimens were obtained to measure bone density,bone calcium and phosphorus contents.Results:Bone densities,bone calcium and phosphorus contents in OVX group at 10 and 20 weeks after treatment were significantly lower than those in sham-operated group and five administration groups(P0.05),bone tissue showed pathological changes of osteoporosis;bone densities,bone calcium and phosphorus contents in estradiol+puerarin 25 group at 10 and 20 weeks after treatment were significantly higher than those in estradiol+puerarin 10 group(P0.05),but compared with sham-operated group,puerarin-50 group,estradiol-200 group and estradiol+puerarin 40 group,there was no significant difference(P0.05).Conclusion:The curative effect of estradiol+puerarin 25 group is superior to that of estradiol+puerarin 10 group,compared with sham-operated group,puerarin-50 group,estradiol-200 group and estradiol+puerarin 40 group,there is no significant difference.
