Study of MG-63 cell growth regulated by exogenous TGF-β1 in vitro

Abstract

Objective To study the effects of exogenous transforming growth factor-β1 (TGF-β1)on regulation of cell cycle progression and expression of the TGF-β1in MG-63human osteosarcoma cells. Methods Human MG-63 osteosarcoma cells were incubated with different concentration of exogenous TGF-β1. The erects of exogenous TGF-β1 on proliferation of human osteosarcoma MG-63 cells were measured with MTT(3-(4,5-dimethylthiazol-2-y1)-2,5-diphenyltetrazoliumbromide) assay. Distribution of cell cycle was studied with flow cytometry with propidium iodide (PI) staining. Induced TGF-β1expression was inhibition of MG-63 cell proliferation which was enhanced by a dose dependent mood. Results Flow cytometry analysis showed that exogenous TGF-β1 blocked the cells mainly in the G1 phase of cell cycle. Expressions of the TGF-β1 in various phases of cell cycle were enhanced. Conclusions Exogenous TGF-β1 can inhibit proliferation of MG-63 cells by the dose depend mood and results in G1 cell cycle arrest. Endogenous TGF-β1 expression of MG-63 cell can be stimulated by exogenous TGF-β1,especially in G0/G1 phase
