Effect of Resveratrol on the Expression of Survivin Protein in Androgen-Independent Prostate Cancer Cell Line PC-3 in Vitro

Abstract

Objective; To investigate the effect of Resveratrol on proliferation inhibition in human pros-tate carcinoma cells PC-3 in vitro and to explore the possible mechanisms involved. Methods; ThePC-3 cells were divided into four treatment groups (25, 50, 100, and 200 £imol/L Resveratrol)and one control group. The morphological alterations of cells PC-3 after treated for different timewere observed by using inverse microscopy; the inhibition of cell proliferation was detected byMTT assay; the protein level of Survivin in carcinoma cells was observed by SP immunohisto-chemistry. Results: After being treated with Resveratrol, PC-3 cells grew round and small obvi-ously and the cell walls were isolated under the inverse microscope; Resveratrol could inhibit theproliferation of PC-3 cells, the inhibition of proliferation of PC-3 cells decreased in a time- anddose-dependent manner, the survival rate of the four groups after 24 hours was 86. 2% , 77.15% ,60.89%, and 35. 76%,respectively, and the rate after 48 hours was 65. 24% , 59.76%, 46.47%,and 27. 77% respectively (P0. 05). Compared with the control group, SP immunohistochemis-try showed the expression of Survivin protein in the PC-3 cells treated with 100 ^mol/L Resvera-trol was decreased (P0. 01). Conclusion; Resveratrol could significantly inhibit the growth ofcells PC-3 in vitro, the induction of apoptosis through down-regulating Survivin protein is proba-bly one of it's molecular mechanisms on inhibition of tumor.
