Effect of Different Sodium Salt Soaking on Seed Germination of Raphanus sativus L. under NaCl Stress

Abstract

[Objective]The aim of the research was to provide references for salt tolerant breeding and cultivation of crops.[Method] The effective method and extent of salt resistance of radish (Raphanus sativus L.) seed was investigated by trial using different sodium salt soaking seeds under NaCl stress. 200 mmol/L NaCl,NaHCO3 and mixed salt (NaHCO3,NaCl) solutions were used in seeds presoaking, then 4 kinds of NaCl solutions with concentrations equally spread from 40 to 160 mmol/L were used in seed germination of radish under NaCl stress. Effect of different sodium salt soaking seeds on seed germination of radish under NaCl stress was analysized through determination of germination percentage, germinative force, vitality index, germination index, lamina fresh weight, lateral root number and chlorophyll content of seedlings.[Result] Mixed salt soaking improved the germination index and vitality index of radish seed, and stimulated the seeding growth significantly. NaHCO3 soaking seed inhibited germination, but improved the number of lateral root and chlorophyll content. The effect of stimulate germination was as follow,mixed salt soaking seedNaCl soaking seedcontrol groupNaHCO3 soaking seed. Under different soaking seed treatment,with the increase of the concentration of NaCl stress solution, the germination was inhibited, while the seeding growth increased at low concentration of NaCl with the increase of the concentration of NaCl stress solution. On contrast, the seeding growth was inhibited with the increase of the concentration of NaCl stress solution when the NaCl concentration was high.[Conclusion] Mixed salt soaking could promote seed germination and seedling growth.
