Effect of lentiviral-delivered PLC-γ1 siRNA on metastasis of human colorectal carcinomas cell lines

Abstract
OBJECTIVE:To investigate the relationship between PLC-γ1 and biological action of coleretal carcinomas by RNA interference recombinant lentivirus that stably suppresses the PLC-γ1 expression in human colorectal carcinomas LoVo cells so that the LoVo cells lines deficient in PLC-γ1 can be established and the role of this gene can be objectively investigated.METHODS:Recombinant lentivirus that produced PLC-γ1 siRNA were prepared.After LoVo cells were transduced with different lentivirust,the stably transduced cells were selected by Blasticidin.the protein and mRNA expression of PLC-γ1 were examined by Western-blot and RT-PCR analysis,and the effect of the lentivirus on the cell metastasis was analyzed by the cell adhesion assay and cell migration assay,respectively.RESULTS:Recombinant lentivirus including PLC-γ1shRNA gene were successfully constructed,PLC-γ1 siRNA knocked down PLC-γ1 expression in LoVo cells obviously.The silenced efficiency of siRNA transducted by recombinant lentivirus was very high.The ability of adhesion and migration of human colorectal carcinomas LoVo cell lines were significantly decreased.CONCLUSION:PLC-γ1 plays an important role in metastasis of human colorectal carcinomas.
