Study of the Level of EGF,PDGF-A and PDGF-B Secreted by Bone Marrow Stromal Cells in Psoriatic Patients

Abstract

Objective To reveal the abnormity of bone marrow microenvironment of psoriasis patients by comparing the level of EGF,PDGF-A and PDGF-B secreted by bone marrow stromal cells in patients and control.Methods Bone marrow mononuclear cells of patients and control by density gradient centrifuge were separated.Bone marrow stromal cells were cultured with the method of adherent.When the cells were cultured for three passages and 72 h,bone marrow stromal cells and supernatant liquid were collected.The phenotypes of cells were identified by flow cytometry and the level of EGF,PDGF-A and PDGF-B were detected by ELISA kits.Results The purity of bone marrow stromal cells was over 90%.The level of EGF and PDGF-B in patients was significantly lower than that in control(P0.05).And the level of PDGF-A was no significant difference between the two groups(P0.05).Conclusion Some of the cytokines were abnormal which secreted by psoriatic bone marrow stromal cells.Which showed that psoriatic bone marrow microenvironment may be abnormal.
