Effect of ischemic post-conditioning on NO expression during renal ischemic reperfusion injury in rat

Abstract

Objective: To observe the change of NO,NOS expression during renal ischemic reperfusion injury in rat after ischemic post-conditioning and to investigate the effect of ischemic post-conditioning on ischemic/reperfusion induced renal injury.Methods: Thirty SD rats were randomly divided into three groups: sham-operated group,ischemic-reperfusion group,ischemic post-conditioning group.The renal underwent 45-minute ischemic and 24-hour reperfusion.The level of Superoxide Dismutase(SOD),Malondialdehyde(MDA),NO,NOS of kidney in each group were examined.the figures of PMN and Paller-grades were counted by hematoxylin-eosin(HE) staining.Results: The levels of Scr,BUN and MDA in ischemic post-conditioning group were significantly lower,while the activities of SOD,NO and NOS were dramatically higher than those in ischemic-reperfusion group.The figures of PMN and Paller-grades were very high in ischemic-reperfusion group while it significantly decreased in ischemic post-conditioning group.Conclusion: The ischemic post-conditioning could protect kidney from ischemic reperfusion injury,it was more likely they were related to the ratios of eNOS and iNOS and the increase of NOS in kidneys.
