A Modified Middle Cerebral Artery Occlusion Method for Establishment of Brain Reperfusion Model in Rats

Abstract

Objective To evaluate the efficacy of a modified method of middle cerebral artery occlusion(MCAO) embolization for establishment of brain reperufusion model in rats.Methods 12 Sprague-Dawley(SD) rats were randomly divided into two groups: control and modification groups,6 rats each.The rats of control group were subjected to temporary middle cerebral artery occlusion(MCAO) by ligation of the pterygopalatine artery(PPA) and insertion of suture-embolus into the external carotid artery(ECA),while the rats of modification group were subjected to temporary MCAO occlusion by insertation of suture-embolus into the common carotid artery(CCA) without PPA ligation.The suture-embolus was removed for reperfursion at 2 hours after occlusion. The neurological scores,operation time,success rate,mortality rate and the infarction ratio in both groups were recorded and analyzed.Results There were no significant differences in neurological scores,success rates,mortality rates and infarct ratios between the two groups.However,the operation time in modification group was significantly shorter in comparison with that in the control group.Conclusion This modified MCAO and reperfusion model by CCA suture-embolus insertation without PPA ligation is a reliable model and worthy of application.
