Study on Extraction and Purification of Hawthorn Leaf Flavonoids

Abstract

Objective:To study the optimal extraction process and purification of hawthorn leaf flavonoids. Methods:Taking the content of general flavonoids asguideline as the index, the preparation technology of hawthorn leaf flavonoids was optimized by using single-factor test and orthogonal test. By using static adsorption,dynamic adsorption and deabsorption of macroporous resin and the purification process were optimized. Results:The results indicated that the optimal process was to extract 10 times 50% ethanol, reflux 3 times, 2 hours for every time. The optimal purification method was to adopt concentration of 0.5 g/mL, herbs and resin ratio 2.4:10(2.4 g herbs:10 mL wet resin),washing the resin with 7BV water to remove impurities,eluting with 6 BV 50% ethanol(elute rate 2 BV/h) through HPD600 macroporous resin, finally obtaining total flavonoids enrichment. The content of total flavonoids content was determined 8.74% in the hawthorn leaf extracts and 85.7% in the purified extracts. Conclusion:The purity of hawthorn leaf flavonoids rises by about ten times through this methods of extraction and purification,providing references for industrial production.
