Abstract
Objective To observe the effect of proliferation inhibition of Arsenic Trioxide (AS_2O_3) on colonic neoplasms cell line SW620 and its mechanism. Methods Colonic neoplasms cell line SW620 was treated with different concentration AS_2O_3. MTT assay was used to observe the proliferation status. Immunohis tochemistry staining method and flow cytometry were used to measure the percentage of proliferating cell nuclear antingen (PCNA) positive cell. Results AS_2O_3 could significantly inhibit the growth of SW620 cell, and this effect was dose and time dependent. The percentage of PCNA positive cell was decreased gradually when concentration of AS_2O_3 was increased. Conclusions Arsenic Trioxide could inhibit the proliferation activity of colonic neoplasms cell line SW620. Reduction of PCNA which was related to the regulation Of DNA synthesis and cell proliferation might be the mechanism of this proliferation inhibition.
