Abstract
Objective To investigate the effect on anti-proliferation of H22 transplanted tumor with different concentration of oridonin and its mechanism in mice.Methods H22 transplanted tumor model was made by injection of H22 tumor cells under skin of the right axillary and high,middle and low dose of oridonin were intragastric administration for 2 weeks.Tumor inhibition rate was measured and the bcl-2,p53 and caspase3 expression were detected by TUNEL and immunohistochemistry.Results H22 transplanted tumor cells were apparently sensitive to oridonin,and oridonin could obviously inhibit tumor proliferation(P0.01).Inhibition rate of high dosage was(48.31±1.74)%,which was nearly same as Mitomycin(P0.05).The positive expression rate of gene bcl-2,p53 and caspase3 were(18.32±0.32)%,(27.57±0.51)% and(62.59±0.57)%.The weight of the mice was not lower down after treatment.Conclusions Oridonin can inhibit the tumor growth in dose-dependent manner,which is related with upregulation the expression of caspase3 and downregulation the expression of bcl-2 and p53 and then activate the apoptosis of the H22 humor cells.
