Abstract
High-speed counter-current chromatography(HSCCC)with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water(5∶5∶7∶7,v/v)was applied to the isolation and purification of alkaloids from Zanthoxylum bungeanum Maxim.The lower phase was used as the mobile phase and performed at a flow rate of 2mL/min,while the apparatus rotated at 800r/min,and detected at 254nm.Two kinds of alkaloids were obtained and yielded 130mg(A),15mg(B)from 800mg of crude extract in a one-step separation.A and B were identified by Thin Layer chromatography(TLC),and the results showed that the two fractions were alkaloids,with the purity of 92.8% and 85.4%,determined by high performance liquid chromatography(HPLC).
