STUDY ON THE EXTRACTION TECHNOLOGY OF POLYSACCHARIDE FROM LYCIUM RUTHENICUM LEAVES

Abstract

Water-bath,microwave assisted and ultrasonic-microwave assisted extraction techniques were applied to extract polysaccharides from Lycium rethenicum leaves and the optimal extraction method was selected by compared with others.Extraction Conditions were assured by a single factor and orthogonal experiments.Content of polysaccharides was determined by anthrone-sulphuric acid colorimetry.The results showed that Ultrasonicmicrowave extraction was the optimum extraction method.The results of an orthogonal experiment indicated that the effectiveness factors on polysaccharides content was the order as follows:temperature extraction time solid-liquid ratio.The technological parameters were as follows:solid-liquid ratio was 1:25,extraction temperature was 90 ℃,extraction time was 30 min.The extraction rate of polysaccharides was 10.07 %,RSD=1.60 %(n=3).Recovery rate was 100.7 %,RSD=1.84 %(n=3).
