Abstract
Objective To observe the effect of oleanolic acid on expression of vascular endothelial growth factor(VEGF) in human leukaemia cancer cell line K562.Methods The inhibitory effect of oleanolic acid on viability of K562 cells was assessed by MTT assay.The expression of VEGF mRNA and protein were determined by RT-PCR and Western blot,respectively.The expression of VEGF in culture medium was detected by ELISA.Results Oleanolic acid inhibited growth of K562 cells in a dose-dependent manner and down-regulated expression of VEGF.Conclusions The anti-tumor mechanism of oleanolic acid is related with inhibition of VEGF expression,which causes down-regulation of VEGF,therefore decreases tumor growth.
