Abstract

Objective: To investigate the effects of different concentration of raloxifene on the proliferation and differentiation of bone marrow stromal cells (BMSCs) from the experimental rats. Methods: Osteoporosis models in Sprague-Dawley (SD) rats were established. BMSCs from the Sham-ovariectomized rats(Sham) and the ovariectomized rats(OVX) were cultured in vitro. BMSCs between passages 3,4 were cultured under different concentration of raloxifene. The proliferation of BMSCs was evaluated by MTT and the differentiation by comparing the synthesized alkaline phosphatase(ALP)was tested by rate method. Results: The 4th passages(P4) BMSCs from the Sham had weaker ability of proliferation than the 3rd passages (P3). But the P4 BMSCs from the OVX had increased ability of proliferation. For the differentiation, there was no statistical difference between the P3 and P4 BMSCs from both group rats. Conclusion: The ability of the proliferation of BMSCs from the Sham dropped quickly after subculturing. The BMSCs from the OVX had the much stronger ability of proliferation and synthesized much more ALP. Raloxifene could promote the growth of the P4 BMSCs from both group rats, but just promoted the differentiation of the BMSCs from the OVX.
