Effect of puerarin on renal expression of fibronectin and type IV collagen proteins in rats with diabetic nephropathy induced by combination of high-fat diet and low-dose streptozotocin

Abstract

Objective: To investigate the effect of puerarin on diabetic nephropathy and the mechanism in rats.Methods: The rat model of diabetic nephropathy was induced by intraperitoneal injection of low-dose of streptozocin(STZ) after having high-fat diet for six weeks.The rats were treated with 3 doses of puerarin,enalapril or XiaoKe Wan every day.At 8 weeks after treatment,biochemical indicators were evaluated.The pathological changes in the kidney were determined by Masson staining.The expression of fibronectin(FN) and type IV collagen(IV-C) were detected by immunohistochemistry.Results: Puerarin suppressed histological changes in the kidney tissues,and reduced the levels of these biochemical parameters and the expression of FN and IV-C proteins in renal tissues of diabetic rats with nephropathy.Conclusion: Puerarin protects renal function and slows down the progression of diabetic nephropathy in rats by suppressing the expression of FN and IV-C proteins in renal tissue.
