Abstract
Objectives to investigate the feasibility and specificity of RNA interference which mediated by SiRNA on the gene therapy for ovarian cancer.Methods The VEGF SiRNA was transfected into OVCAR-3 cells to induce RNAi by using transfecting agent.The changes of VEGF gene expression in both siRNA treatment groups and control groups were measured by RT-PCR,ELASA and Western blotting in vitro.Results Experiments in vitro showed that siRNA directly down-regulated the expression of VEGF in the mRNA and protein level(P 0.05).Conclusions RNAi mediated by siRNA markedly decrease VEGF gene expression and may have the potential as a therapeutic modality to treat human ovarian cancer.
