Abstract
Objective To investigate the effect of Extract of Ginkgo Biloba leaves(EGb50)on NGF mRNA expression after sciatic nerve injury in rats.Methods Sciatic nerve injury model was established by transaction and direct repair of right sciatic nerve in 72 male SD rots,which were randomly divided into two groups.Rats of the experimental group were fed with EGh50(200mg/kg/d)daily while those of the control group fed with normal saline. 6 un-operated rats were employed as normal control.The sciatic nerve distal to the injury site,dorsal mot ganglia and spinal cord were harvested 1,3,7,14,21 and 28 days after the operation and stored in the liquid nitrogen.Total RNA was extracted by the Trizol reagent.Reverse transcripatase-polymerase chain reaction(RT-PCR)was used to detect the mRNA expression of NGF.Results The mRNA expression of NGF in the sciatic nerve of the experimental group was much higher than that of the control group detected on POD 7,14,and 21.7 and 14 days after the operation,the experimental group showed a higher mRNA expression of NGF in dorsal root ganglia and spinal cord than the control group.Conclusion EGb50 can increase the early expression of NGF mRNA in the corresponding dorsal root ganglia and spinal cord of rats sustaining sciatic nerve injury.
