Abstract

【Objective】 To study the inhibition effect of TRAIL combined with Paclitaxel on the osteosarcoma cell lines so as to explore the new method of the clinical treatment.【Methods】 TRAIL and Paclitaxel were used on the OS-732 cells individually or jointly so as to observe their synergistic effects on the apoptosis of osteosarcoma cells.【Results】 Twenty-four hours after the application of 50 ng/mL TRAIL and 10 μg/mL Paclitaxel jointly on OS-732 cells,The inhibition rate is 47.49% by MTT essay which is significantly higher than the application of 50 ng/mL TRAIL(9.85%) and 10 μg/mL Paclitaxel(20.37%) respectively(P 0.01).Morphological changes observed with inverted phase contrast microscope,fluorescence microscope showed tumorkilling effect of joint application was much stronger than respective application.【Conclusion】 Combination of TRAIL and etoposide is obviously conducive to killing effect of osteosarcoma cells which were achieved by inducing apoptosis.
