Matrine derivant X induces apoptosis of human nasopharyngeal carcinoma CNE1 cells by regulation of Caspase-3,Caspase-8 and Caspase-9 expression

Abstract

Aim To study the mechanism of matrine deiwant X inducing apoptosis of human nasopharyngeal carcinoma CNE1 cells in vitro.Methods Transmission electron microscope observation on changes of ultra structure of CNE1 cells was done after CNE1 cells were treated with different dosage of matrine derivant X for 48h,and the protein expression of Caspase-3,-8,-9 was detected by western blotting.Results After incubation of CNE1 cells treated with matrine derivant X for 48h,characteristic morphology of apoptosis appeared,such as cell shrinking,significant increases of in nucleus,increased gap of nucleus periphery,and formation of apoptotic bodies,and the apoptotic characteristics were more visible in high dose group than low dose group.Western blot showed that the expression of Caspase-3,Caspase-8 and Caspase-9 protein increased significantly in a dose-dependent manner in different dosage groups(P0.01) except Caspase-8 in low dosage group compared with negative control group.Conclusion The matrine derivant X can induce apoptosis of CNE1 cells by increasing the Caspase-3,Caspase-8 and Caspase-9 protein expression.
