Detection of Platelet Activation in Coronary Heart Disease with Flow Cytometry

Abstract

Objective:The alpha-granulemembrane protein(CD62P),lysosomal(CD63) were detected in patients with coronary heart disease(CHD) by evaluating the status of in vivo platelet activation of flow cytometry(FCM) and to investigate the relationship between the severity of ocronary artery stenosis and platelet activation.Method:The CD62P、CD63 from whole-blood samples in 90 patients,which including 28 stable angina pectoris(SAP),26 unstable angina pectoris(UAP),16 acute myocardiol infarction(AMI) and 20 angiographically normal were as no CHD controls group.CD62p and CD63 were fluorescently labled with their corresponding monoclonalantibodies(mAb) and analyzed by FCM.Results:There were increases in platelet CD62P and CD63 expression in CHD than those in controls.CD62P and CD63 in the patients with UAP were significantly higher than those in SAP and no CHD.Groups of double and three-vessel disease had significantly increased CD63 or CD 62P comparing with on-vessel disease and singlevessel disease.Conclusion:The platelet activation relates to pathophysiological process of UAP and the severity of ocronary artery stenosis.
