Comparison of pharmacokinetics of chlorogenic acid and constituent chlorogenic acid in Shuanghuanglian Fenzhenji in rats

Abstract
AIM To compare the pharmacokinetics of chlorogenic acid and constituent chlorogenic acid in Shuanghuanglian Fenzhenji in rats.METHODS Rats were intravenously administrated with chlorogenic acid and Shuanghuanglian Fenzhenji containing equal amount of chlorogenic acid in a single dose,respectively.Concentrations of chlorogenic acid in plasma were determined by HPLC-EchD.Main parameters of pharmacokinetics were calculated through DAS software,which were treated by SPSS software for statistics.RESULTS Concentration-time courses of chlorogenic acid alone and chlorogenic acid in Shuanghuanglian Fenzhenji were both fitted to a two-compartment model after intravenous injection.The major pharmacokinetic parameters of chlorogenic acid alone have significant difference from that of chlorogenic acid in Shuanghuanglian Fenzhenji(P 0.05).Compared with group treated with chlorogenic acid alone,the group treated with compoud Shuanghuanglian have obvious increase in ρmax,AUC0-∞ and decrease in CL,Vd.CONCLUSION There are significant difference between pharmacokinetics of chlorogenic acid and constituent chlorogenic acid in Shuanghuanglian Fenzhenji in rats.Other constituents in Shuanghuanglian Fenzhenji have an impact on pharmacokinetics of constituent chlorogenic acid in rats.
