Experimental study of using Octreotide in treatment of acute pancreatitis.

Abstract

Objective To observe changes of serum TNF-α, IL-6 and IL-10 levels in rats with experimental acute necrotic pancreatitis (ANP) treated with Octreotide whereby the therapeutic effects of the drug were evaluated. Methods The acute necrotic pancreatitis (ANP) model was established by injection of 5% sodium taurocholate (0.1 ml/100g) into the common cholangiopancreatic duct in male SD rats. A total of 48 SD rats were equally randomized to three groups: Sham group, ANP group and Octreotide group(30 minutes after inducing the ANP model, the rats received 0. 9 μg Octreotide per 100 g body weight via the femoral vein. Half of the rats studied were sacrificed at 3 hours and the other half at 6 hours in each group after establishment of the study model. The following parameters were determined: scrum amylasc. macroscopic pathologic scoring of the lung and pancreas, pancreatic histological pathologic scoring, the amount of ascitcs and serum levels of TNF-α, IL-6 and IL-10. Results (1)The serum amylase level was significantly increased in ANP groups than that in sham group (P 0. 01). The serum levels of TNF-α, IL-6 and IL-10 in ANP groups were significantly higher than those in sham group with P 0. 001, P 0. 001 and P 0. 001 respectively. (2) In Octreotide group, the serum amylase level, both pancreatic and lung macroscopic pathologic scoring and pancreatic histological pathologic scoring showed no improvement. While the amount of ascites at 6 hours decreased (P 0. 05). The level of serum TNF-α was significantly decreased (P 0.01). There was no change in serum IL-6 and IL-10 levels. Conclusions Octreotide benefits the treatment of acute pancreatitis, possibly by decreasing scrum TNF-αlevel.
