Abstract

Fruiting bodies of Leucocortinarius bulbiger which is Pinus tabulaeformis' Ectomycorrhizal fungi were taken as research material, pure strain was obtained using tissue isolation method. The strain was inoculated in different culture medium, optimum culture medium was selected. The best carbon source, nitrogen source and pH value were selected using the single factor optimization method. The optimum combination of carbon source, nitrogen source and pH value was selected by using multi factor orthogonal experiment. The results indicated that Leucocortinarius bulbiger's best basic culture medium was PACH.On the basis of above result, the optimum carbon source, nitrogen source and pH value of mycelium growing were respectively glucose, ammonium tartrate and pH7.0.Using orthogonal experiments, the three factor combination selected which lead to mycelium growing fast was sucrose, ammonium dihydrogen phosphate and pH 7.0.
