Separation and Purification of Rutoside from Hibiscus Mutabilis Leaves Using High-speed Counter-current Chromatography

Abstract

Objective To develop a experimental system for the separation of rutoside from Hibiscus mutabilis leaves by high-speed counter-current chromatography(HSCCC).Methods HSCCC was successfully applied to purify rutoside extracts of Hibiscus leaves.Firstly,crude extract of Hibiscus mutabilis leaves was prepared by water extraction.Then the crude extract was applied to macroporous resin and eluted with 60% ethanol.The above extract was further sperated by HSCCC.The two-phase solvent system used in HSCCC was ethyl acetate:n-butanol:water(1:0.3:1.5).Results The purity of the obtained rutoside was over 98.6%.Conclusion HSCCC is a feasible method for the purification of rutoside from the herb.
