Abstract
Objective To study the changes and significances of IL-32 in rats with acute pancreatitis(AP) by investigating the changes of IL-32,IL-31,TNF-α and AMY.Methods Sixty-four SD rats were randomly divided into AP group and sham operation(SO) group.Rats both groups were divided into 3h group,6h group,12h group,24h group with 8 in each.AP was induced in rats with retrograde infusion of 4% sodium taurocholate into the the pancreastobiliary duct(1ml/kg).Rats were sacrificed respectively 3,6,12 and 24 hrs after operation.Samples of blood serum and pancreas tissue were collected.AMY in blood serum was detected with automatic biochemistry analyzer.IL-32,IL-31 and TNF-α were determined by enzyme linked immunosorbent assay.The histopathology of pancreas tissues with HE staining were observed under the light microscope.Results 1.Levels of the AMY,TNF-α and pathological score in AP group were significantly higher than those in SO group at the same time(P0.05),and all of them increased gradually with time in the first 6 hrs(P0.05).The level of IL-32 in AP group was significantly higher than that in SO group within 3hrs(P0.01).IL-32 was significantly positively correlated with TNF-a and pathological score within the first 6 hrs in AP group(r=0.899,P0.05,r=0.782,P0.05).IL-32 decreased in AP group 6 hrs later,with no significant difference from that in SO group in exactly 12 hrs.The level of IL-31 in AP group was no different from that in SO group(P0.05) and was not correlated with TNF-α and pathological score at any time.Conclusion IL-32 increases at the early stage of AP and positively correlate with the TNF-α and pathological score.IL-32 may function as a pro-inflammatory factor in the earlier stage of AP.IL-31 has no change in the in the early stage of AP.
