Abstract
Aim: To investigate the effects of implanting allogeneic bone marrow mononuclear cells(BM-MNCs)in the area of acute myocardial infarction on rat ventricular remodeling. Methods:Thirty two male healthy Wistar rats were divided into three groups at random:sham operation group (single thoracotomy,n=8),control group (n=12)and transplantation group (n=12).As soon as the descending anterior branch of left coronary artery was ligated, culture fluid and BM-MNCs fluid were injected into the peri-infarct area of control group and transplantation group, respectively.Four weeks later, changes of left ventricular morphology and cardiac function were evaluated by echocardiography and hemodynamic measurements.Morphologic characteristics of BM-MNCs implanted in peri-infarct areas were observed. Results: Compared with the sham operation group, the left ventricular end-systolic diameter (LVEDs), end-diastolic diameter (LVEDd)and end-diastolic pressure (LVEDP)increased significantly (P0.01), and the left ventricular ejection fraction (LVEF), fractional shortening (LVFS), systolic pressure (LVSP), pressure maximal rates of rise and fall(+dp/dtmax,-dp/dtmax)decreased significantly (P0.01)both in control group and transplantation group four weeks later. LVEDs, LVEDd and LVEDP in transplantation group were lower than those in control group (P0.01) and LVEF, LVFS,+dp/dtmax and-dp/dtmax were higher than those in control group (P0.01). BM-MNCs labeled with BrdU had been observed around the injected sites through immunohistochemical examination. Conclusion: Allogeneic BM-MNCs transplantation improves ventricular remodeling and cardiac function after acute myocardial infarction.
