Abstract
Objective: To observe the effect of genistein on bone microdamage in ovariectomized SD rats at different stages,evaluate the preventive and curative effectiveness of genistein.Methods: Fifty-four SD female rats(aged 10 months) were divided into 3 groups: group A,ovariectomized(OVX);group B,genistein(10 μg/kg every day), and group C(control),At the 4th,8th and 16th week after ovariectomy the rats were killed respectively.The 4th vertebrae were used for fatigue damage tGST by SL22000 bone fatigue experiment device.The parameters of microdamage were measured.Results: Microdamage could be observed in each sample and microcrack was the most usual damaged type.Significant difference in length and number of microcrack per percent of trabecular area was observed(P0.05) between OVX group and genistein group at the 16th week,but not between OVX and control groups at the 4th and 8th week.Significant difference in length and number of microcrack per percent of trabecular area was not observed between genistein and control groups.Conclusion: Genistein could improve bone biological quality at the earlier stage and prevent it from deterioration later.The length and number of microcrack per percent of trabecular area could be used as bone biomechanical marker to evaluate the effectiveness of anti-osteoporostics.
