Abstract
Objective: To investigate the effect of skeletal muscle ischemia/reperfusion injury on the expression of intercellular adhesive molecular (ICAM)-1 in lung. Methods:We established the ischemia/reperfusion model of hind limb in rats. Fifty-four rats were divided into 3 groups: sham operation group(group Ⅰ,n =6),ischemia group(group Ⅱ,n =6),ischemia/reperfusion injury group(group Ⅲ,n =42). The changes of malondialdehyde(MDA) and myeloperoxidase(MPO) in the plasma and the expression of ICAM-1 in lung were determined at different time after operation and reperfusion. Results:In group Ⅲ,both MDA and MPO increased quickly after reperfusion. The expressions of ICAM-1 mRNA and ICAM-1 increased significantly in lung and skeletal muscle soon after reperfusion. ICAM-1mRNA in lung which was detected by using RT-PCR and in situ hybridization reached the peak 12 hours (40.2%±10.8%,P 0.01 vs other groups) after reperfusion,and that of ICAM-1 detected by using immunohistochemistry method reached the peak 12 to 24 hours (43.0%±6.4% and 44.3%±11.8% respectively,P 0.01 vs other teams) and then decreased slowly,still at high level 48 hours after reperfusion. A positive relationship between MDA and ICAM-1 in lung existed. Conclusion:ICAM-1 may play an important role in the damage of lung function after skeletal muscle ischemia/reperfusion injury in rats.
