Abstract

Objective: To investigate the effect of different temperature on the kinetics of lipopolysaccharide( LPS)-induced human THP-1 cell activity in vitro. Methods: Human THP-1 cells were stimulated with LPS at different temperatures( 34℃,37℃ and 40℃),then,TNF-!,IL-1β and NO productions of stimulant supernatant were measured. Also,Levels of TNF-!and IL-1β mR- NA were measured by RT-PCR. Results: Early elevation of TNF-!,IL-1β and NO productions were found in LPS-stimulated THP-1 cells,the highest level was incubated at 40℃,followed by cells that incubated at 37℃ and the lowest level was detected at 34℃. Ex- pression of TNF-!mRNA and IL-1β mRNA were observed as early as 1 h post exposure to LPS in all studied temperatures,but de- creased sharply after 12 h post exposure to LPS in LPS-stimulated THP-1 cell that incubated at 40℃ only. Conclusion: The study de- scribes the striking effects of incubation temperature on activity of LPS-stimulated THP-1 cell.
