Abstract
To study the effect and possible mechanism of thalidomide in treating rat collagen-induced arthritis(CIA).Methods Type Ⅱ chicken collagen was injected into the tails of Spragne-Dewley rats to establish arthritis models.Rats were divided into thalidomide group(10 mg orally per day for 4 weeks), CIA control group,and blank control group.Articular inflammatory index and joint pad thickness were used to evaluate the effect of treatment.Serum tumor necrosis factor-α(TNF-α)level as well as articular pathology index were also measured.Results Four weeks after collagen injection,the articular inflammatory index and joint pad thickness in CIA control group were significantly higher than those in blank control group(P0.001)and improved significantly in thalidomide group(P0.001),while serum TNF-α level and articular pathology index showed no statistically significant difference between thalidomide group and CIA control group.Conclusion Thalidomide is effective in improving inflammation of rat collagen-induced arthritis enroute mechanism other than TNF-α inhibition and it might not inhibit cartilage and bone damage.
