Abstract
Objective To construct recombinant human bone morphogenetic protein 2 (rhBMP2) eukaryotic expressing vector, and to transfect human fibroblasts and other two cell types, and to determine the transient expression of BMP2 in these cells. Methods rhBMP2 eukaryotic expressing vector pRK5 - hBMP2 was constructed and transfected to CHO,NIH3T3 and normal human skin fibroblasts. rhBMP 2 expression was detected with RT - PCR on mRNA level. Results RT - PCR showed that three transfected cells, CHO, NIH3T3 and normal human skin fibroblasts could effectively transcript BMP2 mRNA. Conclusion Recombinant plasmid pRK5 -hBMP2 containing human BMP2 cDNA can transfect human skin fibroblasts and express human BMP2 effectively.
