Abstract
Objective:To investigate the effect of adenosine on cell proliferation and cell cycle of A549 cells.Methods:A549 cells were cultured,the expression of adenosine receptors in A549 cells were detected by RT-PCR.After A549 cells were treated with 0,1,10,100 and 1 000 μmol/L adenosine for 24,48 and 72 hc,ell viability of each group was measured using MTT.After A549 cells were treated with 100μmol/L for 24 hc,ell cycle was analyzed by flow cytometry.One-way ANOVA or independent samples t-test was used to evaluate the difference of all results among different experimental groups.Results:RT-PCR results showed that four different adenosine receptors(A1,A2A,A2B and A3) expressed in A549 cells.After A549 cells were treated with 0,1,10,100 and 1 000 μmol/L adenosine for 24,48 and 72 h,A549 cells survival rate were 91%,84%,80%,73% at 24 h and 82%,69%,58%,55% at 48 h and 86%,56%,52%,38% at 72 h.With the increase of adenosine concentration,A549 cells proliferation were significant inhibited,which showed the concentra-tion dependent effects.Cell cycle analysis revealed a marked increase in the G1 phase(control group:59.07±0.70;100μmol/L:65.05±0.62;t =-11.04,P 0.01)and decrease in the G2 phase(control group:6.67±0.95,100μmol/L:1.29±0.43,t =8.89,P 0.01)following 24 h exposure to 100μmol/L adenosine.Conclusion:Adenosine is able to inhibit the proliferation of A549 cells in concen-tration-dependent and time-dependent manners,and can change the cell cycles.
